Whole-genome sequencing of an in-house isolated Prevotella intermedia strain Pre.in-GSH
Abstract
An anaerobic Prevotella intermedia isolate (strain Pre.in-GSH) was obtained from mouse fecal samples using selective culture-based isolation. Mice were orally administered with 3-methylhistidine (3-MH) for 7 consecutive days, and fecal samples were collected under sterile conditions. Fecal suspensions were serially diluted and cultured on modified PYG agar plates supplemented with 5% sheep blood, 3-MH, hemin, and vitamin K1 under anaerobic conditions. Colonies exhibiting black ring-like halos on blood agar plates were selected and repeatedly purified until single colonies were obtained. The purified isolate was identified as Prevotella intermedia by 16S rRNA gene sequencing and subsequently subjected to whole-genome sequencing to characterize its genomic features and functional potential.
