Extracellular vesicles (EVs) are crucial mediators of intercellular communication that originate from diverse membrane reservoirs. In a genome-wide CRISPR activation screen, Zhong et al. identified MARCKSL1—a membrane adaptor upregulated across many tumor types—as a potentiator of EV secretion from the plasma membrane, but not from mature endosomes.
