Investigation of the Aromatase Cytochrome P450 gene (CYP19) P1.1-PvuII Polymorphism in Native Turkish Cattle Breeds
Abstract
The Aromatase Cytochrome P450 gene (CYP19) encodes the key enzyme in estrogen biosynthesis and has been proposed as a candidate marker for fertility and milk yield traits in cattle. In this study, the CYP19 P1.1-PvuII polymorphism was investigated in five native Turkish cattle breeds. DNA samples obtained from a previous project were used to amplify a 405 bp fragment of the CYP19 gene promoter region, followed by PCR-RFLP analysis. Allele and genotype frequencies were estimated using POPGENE32, and Hardy-Weinberg equilibrium was assessed using a chi-square test. Two alleles (A and B) and two genotypes (AA and AB) were identified among 172 individuals; the BB genotype was not observed. The A allele was monomorphic (1.000) in the Turkish Grey breed and ranged from 0.886 to 0.960 in the remaining breeds. All breeds were in Hardy-Weinberg equilibrium (p>0.05), reflecting a low level of diversity at this locus.
