Evaluation of Early and Remote Antitumor Effects of 1-β-D-arabinofuranosylcytosine and Protons on Ehrlich Carcinoma Ascites Cells Ex Vivo in Mice
Abstract
Abstract The modifying effect of 1-β-D-arabinofuranosylcytosine (AraC) on early and remote antitumor effects in mice exposed to protons in ex vivo conditions was studied. The dependence of the radiosensitizing effect of AraC under the action of protons on the dose of the drug according to the criteria of the antitumor effect in a short term up to 35 days and a long term up to 90 days after exposure is shown. AraC monotherapy enhances median overall survival and lifespan of tumor-bearing mice regardless of concentration. The data obtained in our biomedical model are consistent with current understanding of the molecular mechanisms of induction and repair of potentially lethal DNA damage and their impact on early and late consequences following exposure to repair inhibitors used to enhance proton therapy.
