Sequential skin cancer therapy

Abstract

Background: Skin cancer, particularly melanoma, poses a significant global health challenge due to late diagnosis and treatment resistance. Current therapies often lack personalization and fail to address tumor heterogeneity. Objective: To propose SEQ-SKIN, a sequential integrated therapeutic model that combines cryotherapy, nanomedicine, and immunotherapy to improve outcomes in advanced skin cancer. Methods: We designed a 3-stage sequential protocol: 1. Cryotherapy for initial tumor debulking and antigen release. 2. Nanomedicine-based targeted drug delivery to residual tumor cells. 3. Immune checkpoint inhibitor therapy to stimulate systemic anti-tumor immunity and prevent recurrence. Results: The proposed SEQ-SKIN model leverages synergistic effects between modalities. Cryotherapy enhances antigen presentation, nanoparticles improve drug penetration and reduce toxicity, and immunotherapy provides long-term immune surveillance. This sequence addresses limitations of monotherapies. Conclusion: SEQ-SKIN offers a novel, personalized, and multimodal approach to advanced skin cancer management. It integrates tumor reduction, targeted therapy, and immune activation in a logical sequence to maximize efficacy and minimize resistance. Further clinical validation is warranted.
