SYNTHESIS, SPECTRAL CHARACTERIZATION AND IN VITRO ANTIMALARIAL EVALUATION OF NOVEL QUINOLINE-BASED THIAZOLIDIN-4-ONE DERIVATIVES

Abstract

A series of novel quinoline-based thiazolidin-4-one derivatives were synthesized through a three-step synthetic pathway involving nucleophilic substitution, Schiff base formation and cyclization reactions. Initially, 4-chloroquinoline was converted into 4-hydrazinylquinoline by reaction with hydrazine hydrochloride under reflux conditions. The obtained hydrazine derivative was condensed with various substituted aromatic aldehydes to afford Schiff base intermediates. Finally, cyclization of these intermediates with thioglycolic acid in the presence of anhydrous aluminium chloride produced the desired 2-substituted phenyl-3-(quinolin-4-ylamino)thiazolidin-4-one derivatives. The synthesized compounds were purified by recrystallization and characterized by suitable analytical techniques. The developed synthetic route was found to be simple, efficient and suitable for preparing structurally diverse quinoline-thiazolidinone hybrids.
