Access-Fate-Amplification (AFA): A Compartmental Decision Grammar for Allergic Disease

Central thesis Antigen identity and clinical phenotype do not reconstruct causal architecture. AFA asks which causal object reached which compartment, how genuine non-self was antigen-specifically engaged, which qualitative programme Fate allocated, what state persisted, what exe...

Triple Mechanistic Convergence on MMP-9: A CDP/ACA hypothesis linking aluminum adjuvant, sorbitol, and acetaminophen to early-life neurovascular perturbation and candidate developmental Calibration

Version 1.1 — revision note Version 1.1 integrates the original MMP-9 convergence hypothesis into the Compartmental Decision Pathology (CDP) programme and its neurodevelopmental branch, Access–Calibration–Amplification (ACA). Aluminum adjuvant, parenteral sorbitol, and acetaminop...

The ARA theory of compartmental autoimmunity: Access–Recognition–Amplification as a sequential architecture of autoimmune phenotypes

"Autoimmunity" groups together, under a single term, mechanistically heterogeneous processes that existing models of molecular mimicry and loss of tolerance fail to explain through a shared mechanism. It is proposed that autoimmunity is not a mechanistically homogeneous natural k...

Compartmental Oncogenesis: A Framework of Access, Discrimination, and Amplification in Cancer Development

Central thesis Disease does not begin when pathological potential is generated, but when that potential obtains sustained compartmental access to the conditions that make it consequential. In cancer, the most distinctive form of that access is the one carried out by the altered c...

Microbial Valence Is Not Fixed: A Compartmental Framework for Divergent Autoimmune and Oncogenic Outcomes

Background. Certain microorganisms are associated, in different patients or tissue contexts, with both immune-mediated disease and malignancy. These pairings are usually treated as unrelated associations, as if the microorganism itself carried a fixed pathological sign. Hypothesi...

Periodontal Dysbiosis and Psoriatic Arthritis: From Oral Threshold Modification to Post-Enthesitic Synovial Amplification

Abstract Background Psoriatic arthritis (PsA) is not well explained by directly importing the Porphyromonas gingivalis–PPAD–ACPA model from rheumatoid arthritis (RA): PsA is usually seronegative and has an enthesis-centered anatomy. This note develops a distinct hypothesis: chron...

Cumulative Inflammatory Phagocytic Reprocessing of Persistent Particulate Aluminum

A phagocyte that engulfs a persistent particulate load does not neutralise it: the cell may be activated, injured or replaced, but the material itself survives, and what happens next depends on which cell takes it up in its place. This hypothesis paper proposes a multiexposure–mu...

Excipient-Mediated Vectorisation Across Biological Barriers: The Vascular Escape–Corona–Transcytosis (VECT) Model of Iatrogenic Disease

Abstract Background Biological barriers are selectively permeable interfaces whose receptor-mediated transport systems create a structural vulnerability: receptors recognise ligands on the exterior of approaching complexes and initiate transcytosis without evaluating the full int...

Streptococcus pyogenes and Sydenham Chorea: Molecular Antigenic Mimicry with Barrier-Dependent Neuroimmune Access

Abstract Background Other documents in this series have developed vascular access and lesion topology, and cellular second-hit trafficking of adjuvant particles, as convergent but mechanistically distinct routes to compartmental autoimmune or post-infectious disease, within the A...

Vascular and Leukocyte-Mediated Access in Bacterial Neurological Disease: A Lesion-Topology Framework for Phenotypic Divergence

Abstract Background Rickettsia rickettsii/conorii is an endotheliotropic bacterium with a demonstrated capacity to activate human vascular endothelium via NF-κB-dependent pathways, a mechanism also reported, with more limited evidence, in Treponema pallidum [1-4]; Ehrlichia chaff...
